Some requirements for pyruvate oxidation by plant mitochondrial preparations.

چکیده
coenzyme A becomes progressively less under these conditions. As previous authors have pointed out (e.g. Laties, 1953b; Lindberg & Ernster, 1954) such a result might be expected if a single dehydrogenase well supplied with substrate was sufficiently active to saturate an electron-transfer system normally shared by subsequent dehydrogenation steps. In conclusion it may be mentioned that responses to coenzyme A and cocarboxylase may be expected when other preparations are investigated, although the high rates of oxygen uptake which have been recorded without benefit of such additions may indicate a degree of self sufficiency which would make these preparations less desirable for such demonstrations. However, we have already been able to show responses to these cofactors in the oxidation of oxaloacetate by preparations from cauliflower and potato.

Effect of chloral hydrate and acetaldehyde on mitochondrial preparations from sweet potato.
The inhibitory effects of chloral hydrate and acetaldehyde have been studied on oxidations performed by mitochondrial preparations of sweet potatoes (Ipomea batatas). With a variety of substrates, chloral acts very like amytal but only between a fifth and a tenth as effectively; it affects those reactions which would be expected to depend on the oxidation of intramitochondrial DPNH, more than t...
متن کاملIsolation and oxidative properties of intact mitochondria isolated from spinach leaves.
A procedure was described for preparing intact mitochondria from spinach (Spinacia oleracea L.) leaves. These mitochondria oxidized succinate, malate, pyruvate, alpha-ketoglutarate, and NADH with good respiratory control and ADP/O ratios comparable to those observed with mitochondria from other plant tissues. Glycine was oxidized by the preparations. This oxidation linked to the mitochondrial e...
متن کاملIsolation and Oxidative Properties of Intact Mitochondria from the Leaves of Sedum praealtum: A Crassulacean Acid Metabolism Plant.
A procedure is described for preparing intact mitochondria from leaves of Sedum praealtum D.C., a plant showing Crassulacean acid metabolism. These mitochondria oxidized malate, pyruvate, alpha-ketoglutarate, succinate, NADH, NADPH, and isocitrate with good respiratory control and ADP/O ratios better than those observed in mitochondria from other photosynthetic tissues.Malate oxidation was very...
متن کاملCofactor Requirements for Oxidation of Alpha-Keto Acids by Sweet Potato Mitochondria.
In recent years evidence that suggests the operation of the Krebs cycle in plant tissues has accumulated. Studies by Millerd et al (10), Laties (8), Brummond and Burris (4), Biale and Young (3), and Smillie (14), have demonstrated that mitochondria, isolated by differential centrifugation of tissue homogenates, can carry out oxidation with concomitant phosphorylation of Krebs cycle acids. Davie...
متن کاملSpecificity of the Organic Acid Activation of Alternative Oxidase in Plant Mitochondria.
The claim that succinate and malate can directly stimulate the activity of the alternative oxidase in plant mitochondria (A.M. Wagner, C.W.M. van den Bergen, H. Wincencjusz [1995] Plant Physiol 108: 1035-1042) was reinvestigated using sweet potato (Ipomoea batatas L.) mitochondria. In whole mitochondria, succinate (in the presence of malonate) and both L- and D-malate stimulated respiration via...
